Keep a mini medicine/hygiene kit in your diaper bag
Amy passed along a tip for keeping the diaper bag (relatively) organized:
It seems like we can never find what we're after in our diaper bag, so we keep a small zippered case (I think ours was originally a brush/comb/nail clipper/etc. set, but any small pencil case or make up bag would do) in the diaper bag that is for Tylenol, Ibuprofen, gas drops, a nail file, a nasal aspirator and other small medicine/hygiene items like that. It makes the small items easy to find in the cavernous bag, and makes it easy to check and see if we need a refill before we leave the house.
[I used to keep a full change of baby clothes in a ziploc bag inside the diaper bag -- same idea -- it was easy to grab if we had a blowout away from home. -- Ed.]

That's a great tip. If your diaper bag has enough compartments you could just relegate all supplies into that particular section too.
